Chromogranin can be used to with calretinin, inhibin and melan-A to distinguish adrenal cortical lesions (chromogranin -, calretinin+, inhibin+, melan-A+) from adrenal medullary lesions (pheochromocytomas; chromogranin+, calretinin-, inhibin-, melan-A-).  Synaptophysin is not as specific as chromogranin when determining the origin of adrenal tumors (cortical vs medullary) since synaptophysin can stain adrenal cortical tumors as well as medullary tumors.
Chromogranin can be used with synaptophysin and CD56 to identify neuroendocrine tumors.
